no, you need to do it from an x86_64 machine.
You can make i[3,4,5,6]86 RPMS on an x86_64 machine ... can't do x86_64 RPMS from a i686 machine.
When I tried to do so with this command
rpmbuild -ba --target x86_64 kernel-2.6.spec
I got the following error:
