The new yum can handle a normal proxy via the mirrorlist selection
process (the non-centos version of yum 2.4 can not). It can also handle
a transparent squid proxy ok most of the time ... though you will have
issues when doing a major upgrade as some of the cached stuff is wrong
if you hit the server in the middle of the update process.

It does not handle the MS ISA server very well.
